dc.description.abstractPour contourner les limites du criblage expérimental, une nouvelle approche alternative basée principalement sur des techniques informatiques a été envisagée : c’est le docking moléculaire. Cette approche permet de modéliser les interactions entre une protéine et des milliers de candidats moléculaires. Dans notre étude, nous avons utilisé le programme Molegro afin de rechercher de nouveaux inhibiteurs de la MAO-B ; cible thérapeutique validée pour le traitement de la maladie de parkinson. L’amarrage moléculaire de la pipérine et ses 26 analogues structuraux issus de sa modification structurale fait ressortir les composés : L7, L16, L17, L18, L20, L21 et L22 comme nouveaux inhibiteurs de la MAO-B avec les scores respectifs de -143.03,-143.545,-150.022, -152.76, -141.771, -145.454 et -154,128 KJ/Mol. L’application de la règle de Lipinski, la vérification de la solubilité dans l’eau, l’accessibilité à la synthèse, les paramètres pharmacocinétiques, ainsi que les tests de toxicité potentielle nous renseignent de manière positive sur les propriétés ADME/tox de ces nouvelles molécules. To overcome the limitations of experimental screening, a new alternative approach based mainly on computer techniques has been envisaged: molecular docking. This approach makes it possible to model the interactions between a protein and thousands of molecular candidates. In our study, we used the Molegro program to search for new inhibitors of MAO-B, a validated therapeutic target for the treatment of Parkinson's disease. The molecular docking of piperin and its 26 structural analogues resulting from its structural modification highlights the compounds: L7, L16, L17, L18, L20, L21 and L22 as novel MAO-B inhibitors with respective scores of -143.03, -143.545, -150.022, -152.76, - 141.77,-145.454 and -154.128 KJ/Mol.Application of Lipinski's rule, verification of water solubility, accessibility to synthesis, pharmacokinetic parameters and potential toxicity tests provide positive information on the ADME/tox properties of these new molecules.en_US
